What Matthew 21:43 means
Based on their rejection of the Son, Jesus solemnly declared, "The kingdom of God shall be taken away from you, and shall be given to a nation bringing forth the fruits thereof." This pronounced a divine judgment upon the corrupt religious leadership of Israel, indicating that the privilege and responsibility of bearing spiritual fruit in God's Kingdom would be transferred from them to those, both Jew and Gentile, who would embrace Christ and live in obedience.

Context
This verse delivers Jesus' explicit and profound judgment, directly applying the parable of the wicked tenants and the prophecy of the rejected stone to the chief priests and elders. It marks the formal declaration of the transfer of the Kingdom of God, immediately preceding a further warning about the consequences of rejecting Him. This statement is a stark climax, exposing the spiritual fate of His adversaries.
v.42Jesus saith unto them, Did ye never read in the scriptures, The stone which the builders rejected, The same was made the head of the corner; This was from the Lord, And it is marvellous in our eyes?
